OGG1 rs1052133 (Ser326Cys) POLYMORPHISM AND URINARY 8-oxodG LEVELS IN PATIENTS WITH RELAPSING-REMITTING MULTIPLE SCLEROSIS: A CASE-CONTROL STUDY /

Abstract

 

 

Background: Oxidative stress is considered an important contributor to the pathogenesis of multiple sclerosis (MS). 8-oxo-7,8-dihydro-2’-deoxyguanosine (8-oxodG) is a well-established biomarker of oxidative DNA damage, while 8-oxoguanine DNA glycosylase 1 (OGG1) is a key enzyme involved in base excision repair. The common OGG1 rs1052133 (Ser326Cys) polymorphism has been associated with altered repair efficiency in oxidative stress. Aim: To investigate the association between OGG1 rs1052133 polymorphism and urinary 8-oxodG/creatinine levels in patients with relapsing-remitting MS (RRMS). Methods: This case-control study included 152 RRMS patients and 247 healthy controls. Genotyping of OGG1 rs1052133 was performed using TaqMan real-time PCR. Urinary 8-oxodG was measured by HPLC-MS/MS and expressed as the 8-oxodG/creatinine ratio (nmol/mmol). Genotype distributions were tested for Hardy-Weinberg equilibrium. Differences between groups were analyzed using the λ2 test and ANOVA with post hoc Sidac test. Results: Genotype and allele distributions did not differ significantly between MS patients and controls (p > 0.05). MS patients showed significantly higher urinary 8-oxodG/creatinine levels compared to controls. Among CC and CG genotypes, MS patients had significantly higher levels than corresponding controls (p=0.005 and p < 0.001, respectively), whereas no difference was observed in GG homozygotes. Within the MS group, carriers of the G allele (CG+GG) had higher 8-oxodG levels compared to CC homozygotes (p=0.029). Conclusion: OGG1 rs1052133 was not associated with MS susceptibility in the studied population. However, the polymorphism appears to modulate oxidative DNA damage levels in MS, suggesting a potential genotype-dependent effect on oxidative stress response. Увод: Оксидативни стрес се сматра важним фактором у патогенези мултипле склерозе (МС). 8-оксо-7,8-дихидро-2´-деоксигванозин (8-оксодГ) представља добро утврђен биомаркер оксидативног оштећења ДНК, док је 8-оксогванин ДНК гликозилаза 1 (ОГГ1) кључни ензим укључен у поправку база путем механизма ексцизионе репарације. Уобичајени полиморфизам ОГГ1 рс1052133 (Ser326Cys) повезан је са измењеном ефикасношћу репарације у условима оксидативног стреса. Циљ: Испитати повезаност између ОГГ1 рс1052133 полиморфизма и нивоа 8-оксодГ/креатинин у урину код пацијената са релапсно-ремитентном мултиплом склерозом (РРМС). Методе: Студија типа случај-контрола обухватила је 152 пацијента са РРМС и 247 здравих контрола. Генотипизација ОГГ1 рс1052133 извршена је применом TaqMan real-time PCR методе. Ниво 8-оксодГ у урину одређен је методом ХПЛЦ-МС/МС и изражен као однос 8-оксодГ/креатинин (нмол/ммол). Расподела генотипова тестирана је на Hardy-Weinbergovu  равнотежу. Разлике између група анализиране су λ2 тестом и АНОВА анализом уз пост хок Сидак тест. Резултати: Расподела генотипова и алела није се значајно разликовала између пацијената са МС и контролне групе (п > 0,05). Пацијенти са МС су имали значајно више концентрације 8-оксодГ/креатинин у урину у поређењу са контролама. Код ЦЦ и ЦГ генотипова, пацијенти са МС имали су значајно више вредности у односу на одговарајуће контроле (п=0,005 и п<0,001), док код ГГ хомозигота није уочена разлика. Унутар МС групе, носиоци Г алела (ЦГ+ГГ) имали су више нивое 8-оксодГ у поређењу са ЦЦ хомозиготима (п=0,029). Закључак: ОГГ1 рс1052133 није био повезан са подложношћу за МС у испитиваној популацији. Међутим, овај полиморфизам може утицати на ниво оксидативног оштећења ДНК код МС, указујући на потенцијални генотип-зависни ефекат у одговору на оксидативни стрес.
